Allopurinol-riboside competitively inhibits the action of purine nucleoside phosphorylase on inosine with a Ki of 277 μM. Lymphocyte blastogensis induced by PHA and Con A is significantly suppressed by allopurinol-riboside in a concentration-dependent manner. When LPS is used as a mitogen, the inhibition of allopurinol-ribosideon lymphocyte proliferation is less marked. Humoral immunity is not suppressed by allopurinol-riboside[1]. Allopurinol riboside is an experimental agent for the treatment of leishmaniasis and American trypanosomiasis. Allopurinol riboside is effective against parasites, because a series of enzymes (analogous to those that mediate purine salvage in humans) convert it into 4-aminopyrazolopyrimidine ribonucleoside triphosphate, a cytotoxic product. Allopurinol riboside is selectively toxic, because it is not metabolized by the corresponding enzymes in humans[2].

Allopurinol riboside peaks in plasma 1.6 hours after administration, has an elimination half-life of 3 hours, and steady-state concentrations in the therapeutic range[3]. After oral administration, unexpectedly low levels of allopurinol riboside in plasma are attributable to incomplete absorption and rapid renal clearance. Probenecid reduces the renal clearance of allopurinol riboside, extends the half-life of allopurinol riboside in plasma, and triples the levels of allopurinol riboside in plasma[4].

Nicotinamide riboside Chloride, an orally active NAD+ precursor, increases NAD+ levels and activates SIRT1 and SIRT3. Nicotinamide riboside Chloride is a source of vitamin B3 (niacin) and enhances oxidative metabolism, protection against high fat diet-induced metabolic abnormalities[1]. Nicotinamide riboside Chloride reduces cognitive deterioration in a transgenic mouse model of Alzheimer’s disease[2].

Nicotinamide riboside Chloride increases intracellular and mitochondrial NAD+ content in C2C12, Hepa1.6, and HEK293 cells in a concentration-dependent manner at concentrations ranging from 1-1000 μM[1].
Nicotinamide riboside chloride boosts NAD to restore antiviral poly(ADP-ribose) polymerase (PARP) functions to support innate immunity for coronavirus (CoVs), a cause of COVID-19[3].

Adenosine affects almost all aspects of cellular physiology, including neuronal activity, vascular function, platelet aggregation, and blood cell regulation[1][2].

Adenosine (Adenine riboside) acts on four G-protein coupled receptors: two of them, A1 and A3, are primarily coupled to Gi family G proteins; and two of them, A2A and A2B, are mostly coupled to Gs like G proteins. These receptors are antagonized by xanthines including caffeine. Via these receptors it affects many cells and organs, usually having a cytoprotective function[2].
Adenosine is an extracellular signaling molecule that is generated from its precursor molecules 5’-adenosine triphosphate (ATP) and 5’-adenosine monophosphate (AMP)[3].
Adenosine is a common metabolite of ATP, which exhibits cytotoxic effects at high concentrations.Adenosine (1.0-4.0 mM; 12-24 hours) inhibits cell viability and triggers ER stress in HepG2 cells[4].
Adenosine induces apoptosis in a variety of cancer cells. Adenosine (2.0 mM; 12-24 hours) induces autophagy in HepG2 cells. In HepG2 cell lines, Adenosine -induced AMPK/mTOR pathway activation partially blocked ER stress and decreased apoptotic cell death[4].
