◆INDIGO的产品平台


INDIGO Biosciences的细胞核受体检测产品是一种基于细胞的报告基因检测系统，其特点是使用INDIGO的CryoMite™工艺来制备工程受体特异性报告细胞。复苏后，报告细胞的活性通常会超过95%，并且可立即使用。无需进行离心-洗涤、活性检测或细胞滴度调整等中间步骤。待测化合物可通过与健康、正在分裂的哺乳动物细胞的细胞质和核环境所表达的核受体反应来进行激动剂或拮抗剂活性的筛选。

INDIGO的产品利用了生物发光报告基因技术的低背景、高灵敏度和广泛的线性动态范围的特点。报告细胞中所含的cDNA编码的甲虫荧光素酶，是一种北美萤火虫（Photinus pyralis）来源的62 kDa蛋白。荧光素酶在Mg2+ 依赖性反应中消耗O2 和ATP作为共底物，催化D-荧光素的单氧化，并产生氧化荧光素、AMP、PPi、CO2 和光子发射产物。反应的发光强度，使用光度计定量并以相对光单位（RLU）进行报告。因此，对经待测样品处理过的报告细胞中的荧光素酶的表达进行定量检测，提供了一种可替代活性变化的灵敏检测方法。 

INDIGO的报告细胞系统经过精心设计，在定量受体活性时，可提供理想的检测灵敏度和动态范围。随着配体的激活，受体复合物起到诱导荧光素酶报告基因表达的作用。添加检测试剂后，荧光素酶反应的发光强度与特定受体的激活状态直接相关。所有INDIGO受体报告基因检测系统均含有专门配制的检测试剂，可在荧光素酶反应开始后5- 90 min内提供稳定的光发射，因此，在开始活性检测前，可将检测试剂分装至所有检测孔中。这不仅免除了对配备进样器的光度计的需要，还可以分批处理孔板，从而大幅度减少了检测板从开始到结束的读取时间。

INDIGO Biosciences的核受体报告基因检测系统经过配置优化，可在96孔和384孔型的检测板中进行处理。单板试剂盒（1×96孔和1×384孔）以全包含式检测系统的形式提供。各试剂盒包含核受体报告细胞、1个检测板、所有必需的试剂（优化培养基、阳性对照激动剂和检测试剂），以及详细的技术手册。

另可提供大包装试剂包，以满足HTS（高通量筛选平台）用户的需求。这些产品适用于10×96和10×384孔板分析，包含核受体报告细胞、优化培养基、阳性对照配体、荧光素酶检测试剂以及详细的技术手册和方法快速指南。由于HTS设备通常储存着大量的检测板，因此散装试剂包中不含检测板。

请注意，无论何种产品规格，产品中的核受体报告细胞均为一次性试剂。一旦复苏，报告细胞就不能再冷冻或重复使用。检测试剂盒提供了足量的试剂，可满足执行机械转移和待测化合物稀释的需要。但每次进行检测设置后，应丢弃多余的报告细胞和荧光素酶检测试剂。

INDIGO的核受体报告基因检测系统试剂盒有以下五种基本规格：

●　96孔板、3×32孔型

此类全包含式试剂盒包含报告细胞和足够的材料，可进行三组不同的32次检测。每组包括32个核受体检测，利用4个8孔条固定的96孔框（已提供）。如有需要，可将试剂同时以64次或全部96次检测反应进行组合。

●　1×96孔型

此类全包含式试剂盒包含报告细胞和所需的其他全部材料，适合需要将整个96孔板用于核受体检测的用户。

●　1×384孔型

此类全包含式试剂盒适合需要将整个384孔板用于核受体检测的用户。

●　（10×）96孔板、960次检测大包装试剂包

此类试剂盒可用于HTS设备，包含至少10×96孔板用的实验材料。本产品不提供检测板。因此，用户必须自行准备白色、无菌、适合细胞培养用的96孔检测板。

●　（10×）384孔板、3,840次检测散装试剂包

此类试剂盒可用于HTS设备，包含至少10×384孔板用的实验材料。本产品不提供检测板。因此，用户必须自行准备白色、无菌、适合细胞培养用的384孔检测板。
