PMQA (Zn-green), an 8-aminoquinoline-based ratiometric fluorescent sensor, demonstrates the Zn2+-induced redshift of emission (85 nm). PMQA (Zn-green) is a cell membrane-permeable probe and suitable for imaging Zn2+ in living cells[1].

After incubation with PMQA (20 μM) for 1 h, weak green fluorescence appeared, presumably caused by the probe capturing intracellular zinc from its native ligands since PMQA has high affinity for zinc[1]. The response of PMQA to Zn2+ can be reversed by N,N,N’,N’-tetrakis(2-pyridylmethyl)ethylenediamine (TPEN) or EDTA[1]. When the stock solution of PMQA (0.1 mM in Tris Buffer) is kept at room temperature for a week, there is no significant loss of PMQA determined by HPLC analysis[1].

NEP (VDP-green (NEP)) is a turn-on fluorescent probe based on the intramolecular charge transfer (ICT) mechanism for sensing vicinal dithiol-containing proteins (VDPs). NEP exhibits high selectivity toward VDPs in live cells and in vivo and displays a strong green fluorescence signal (λex/λem=430/535 nm). NEP has the potential for parkinsonism[1].

NEP (1-30 μM; 6 hours) has no or little cytotoxicity in HepG2 and PC12 cells[1]. NEP (10 μM; 4 hours) causes the fluorescence intensity to decrease gradually in PC12 cells pretreated with 6-OHDA (0, 50, 100, and 200 μM; for 30 min)[1]. NEP contains a dithiarsolane moiety (five-membered As-S ring) as the receptor of VDPs. In the presence of VDPs, NEP displays a strong green fluorescence signal produced by the cyclic dithiarsolane cleavage and subsequent intramolecular cyclization to liberate the fluorophore. NEP maintains a reliable fluorescence response within the range of pH 7-10[1].

AC-green (VDP-green) is a β-allyl carbamate fluorescent probe for specifically imaging vicinal dithiol proteins (VDPs) in living systems (λex/λem=400/475 nm). AC-green can detect the reduced bovine serum albumin (rBSA) with high sensitivity. AC-green displays low toxicity and features high sensitivity, and is suitable for sensing VDPs in living cells and zebrafishes[1].

AC-green (VDP-green) can respond to VDPs with more than 60-fold increase of emission in aqueous solution, while there is no significant interference from biological thiols, amino acids or inorganic salts[1]. AC-green (2 μM; 90 min) has no apparent fluorescence signal within the pH range of 5.0-9.0. Addition of rBSA turns on the fluorescence[1]. AC-gree (10 μM) has low cytotoxicity in HepG2 cells and Hela cells[1]. AC-gree (10 μM; for 15 min) images VDPs in living HepG2 cells and bright green fluorescence appeared. This fluorescence is inhibited when the cells are pretreated with PAO, a popular specificligand for protein vicinal dithiols[1]. AC-gree (10 μM; for 20 min) incubates with zebrafishes has a strong fluorescence signal appeared in the green channel[1].

Malachite green oxalate is also a potent and selective inhibitor of IKBKE, and inhibits its downstream targets such as IκBα, p65 and IRF3. Malachite green oxalate exhibits antitumor activity in vitro and in vivo[1][2][3].

Msr-Ratio (Msr-green) is a ratiometric fluorescent probe of methionine sulfoxide reductase (λex=375 nm, λem=550 nm). Msr-Ratio is used for monitoring the enzyme activity in vitro and in live cells[1].

Msr-Ratio is converted to its corresponding sulfide form by methionine sulfoxide reductase. Msr-Ratio displays favorable properties such as a nearly 400-fold fluorescence change, fast response rate (<30 min), large stokes shift (120 nm), and green emission (550 nm)[1]. There is a very weak background signal in live HL60 cells after the addition of Msr-Ratio. As the Msr-Ratio (10 μM) incubation time increased in cells from 0 to 8 h, the fluorescence signal constantly strengthened[1].
