(-)-Epigallocatechin Gallate is a tea flavonoid with potent antioxidant, antiinflammatory, and anticarcinogenic properties. (-)-Epigallocatechin Gallate is reported to inhibit EGFR signaling and thereby exert anticancer effects[1]. (-)-Epigallocatechin Gallate (EGCG) is a glutamate dehydrogenase 1/2 (GDH1/2, GLUD1/2) inhibitor. Epigallocatechin-3-gallate induces oxidative phosphorylation (OXPHOS) by activating cytochrome c oxidase.

(In Vitro)

(-)-Epigallocatechin Gallate (EGCG) inhibits activation EGFR, HER2 and HER3 in the SW837 human colon cancer cell line. (-)-Epigallocatechin Gallate (10 μM) also inhibits cell growth, suppresses activation of EGFR, HER2, and HER3, and causes decrease in the levels of COX-2 and Bcl-xL proteins, and apoptosis after treatment for 96 h[1]. (-)-Epigallocatechin Gallate (0-35 μg/mL) inhibits the proliferation of colorectal cancer cells. (-)-Epigallocatechin Gallate (35 μg/mL) induces apoptosis of colorectal cancer cells[3]. (-)-Epigallocatechin Gallate (EGCG; 50, 75 and 100 μM) dose-dependently inhibits the growth of HepG2 cells, and induces apoptosis in HepG2 cells[4].

LoVo, SW480, HCT-8, and HT-29 cells are seeded in 96-well plates at a concentration of 5×103 cells; each cell line is totally seeded in the 12 wells. Complete medium is added to the wells, up to 200 μL; the medium contains 0 μg/mL, 10 μg/mL, 20 μg/mL, and 35 μg/mL of (-)-Epigallocatechin Gallate. The inhibition rate=[1 – (absorbance of (-)-Epigallocatechin Gallate group – absorbance of control group)/(absorbance of control group – absorbance of blank control group)] × 100[3].

Mice[3]
At 2 weeks postsurgery, 39 out of the 40 nude mice presented with tumors. Based on the volume of the tumors, the 39 mice with tumors are divided into four groups: a control group (n=9); a group that receives 5 mg/kg of (-)-Epigallocatechin Gallate (n=10); a group that receives 10 mg/kg of (-)-Epigallocatechin Gallate (n=10); and a group that receives 20 mg/kg of (-)-Epigallocatechin Gallate (n=10). In the therapeutic groups, (-)-Epigallocatechin Gallate is administrated intragastrically, and in the control group, 100 uL of physiological saline is administrated intragastrically, once daily for 14 days. After the treatment of the mice with (-)-Epigallocatechin Gallate for 4 weeks, the growth and metastasis of the primary tumors are continuously monitored using a fluorescent imaging system. After 4 weeks, the primary tumors are weighed and immediately put into liquid nitrogen (−196°C) and 2 to 3 hours later, these specimens are stored at −80°C. In addition, the other parts of the primary tumor and metastases are fixed in 4% formaldehyde[3].

(-)-Epicatechin gallate (Epicatechin gallate), a component of Rhei Rhizoma, is one of the active components of Onpi-to, a herbal medicine composed of five crude drugs (Rhei Rhizome, Glycyrrhizae Radix, Ginseng Radix, Zingiberis Rhizoma and Aconiti Tuber). Following intravenous injection of (-)-Epicatechin gallate (1.0 mg/kg) in rats, the plasma concentration vs. time curve is fitted in a three compartment model. Pharmacokinetic parameters for plasma Epicatechin gallate (ECG) are measured. ECG has a t1/2α of 0.038 h, a t1/2βof 0.291 h and a t1/2γ of 4.033 h . The CLtot of ECG is 4.19 L/h • kg. The Vdss is 12.39 L/kg[2].

Animal Administration

Rats[2]
Male Sprague-Dawley rats, obtained at 7 weeks of age (210-245 g) are used. Epicatechin gallate (ECG) is suspended in 0.5% w/v sodium carboxymethylcellulose at 12.5, 25.0 and 50.0 mg/10 mL for oral administration to rats at 10 mL/kg. For intravenous injection in rats at 1.0 mg/kg, an ethanolic solution of Epicatechin gallate is diluted with 10% w/v sodium citrate solution to 1.0 mg/mL; the final concentration of ethanol is 1% v/v[2].

(−)-gallocatechin gallate in combination with active catechins ((−)-epigallocatechin gallate) has synergistic effects on the induction of apoptosis and inhibition of cell growth for PC-9 cells. (−)-gallocatechin gallate shows inhibitory effect on α-Glucosidase and DPPH, with IC50s of 30.2 μM and 12.2 μg/mL[2].

Cell Assay

Cell viability is examined by measuring the capability of cells to metabolize MTT to a purple formazan dye. Human liver cancer HepG2 cells are maintained in DMEM medium supplemented with 10% fetal bovine serum, 100 units/mL penicillin, and 50 units/mL streptomycin at 37°C in a humidified incubator with 5% CO2 atmosphere. Cells are seeded in 96-well tissue culture plates for 24 h and then incubated with the tested compounds at different concentrations for 72 h. After incubation, 25 μL MTT in 5 mg/mL PBS is added and incubated for 4 h. The medium is aspirated and replaced with 150 μL dimethyl sulfoxide (DMSO) to dissolve the formazan salt. The color intensity of the formazan solution, which reflects the cell growth condition, is measured at 570 nm using a microplate spectrophotometer.

(-)-Catechin gallate (CG) directly interacts with DNA oligomers and inhibits the activity of COX-1 and COX-2 enzymes, the gene expression of matrix metalloproteinase-9 in macrophage-differentiated HL-60 myeloid leukemia cells, the adipocyte uptake of glucose by the transporter, GLUT4, and the activities of various proteasomes, i.e., the multicatalytic proteases responsible for the degradation of most cellular proteins. The relative cytotoxicities of a 3-day exposure to (-)-Catechin gallate are determined for cancerous CAL27 and HSG cells, immortalized epithelioid S-G cells, and normal HGF-1 gingival fibroblasts. The concentration at which toxicity (P≤0.01) initially occur is 25 μM (-)-Catechin gallate for S-G cells, 50 μM (-)-Catechin gallate for CAL27 cells, 62.5 μM (-)-Catechin gallate for HSG cells and 75 μM (-)-Catechin gallate for HGF-1 fibroblasts. The calculated neutral red (NR50) values for a 3-day exposure to (-)-Catechin gallate are 58 μM for S-G cells, 62 μM for CAL27 cells, 90 μM for HSG cells and 132 μM for HGF-1 fibroblasts[1].

Cell Assay

Human tongue squamous carcinoma (CAL27) cells and human salivary gland carcinoma (HSG) cells are used. Individual wells of a 96-well microtiter tissue culture plate are inoculated with 0.2 mL of the growth medium containing 2×104 cells/well for a 1-day exposure, 1.5×104 cells/ well for a 2-day exposure and 1×104 cells/well for a 3-day exposure to the test agents. After 1 day of incubation, the growth medium is removed and replaced with exposure medium, with or without varied concentrations of the test agents. In some studies the cells are coexposed to (-)-Catechin gallate (100, 200, 300, 400, and 500 μM) and 100 Units/mL catalase. After 1-3 days of exposure to the test agents, viability is assessed with the neutral red (NR) assay, which is based on the uptake and accumulation of the supravital dye, neutral red (NR)[1].
