It is found that Autocamtide-2-related inhibitory peptide (a novel synthetic peptide ATP) is a potent inhibitor of CaM-kinase II with an IC50 of 40 nM, which is active in the presence or absence of Ca2+/calmodulin. It is 50 and 500 times more potent than CaMK-(281-302Ala286) and KN-93,respectively, under the assay condition used[1].

Eledoisin Related Peptide shares with Substance P (SP) a common N-terminal amino acid sequence and has been shown by to have SP-like activity in the periphery (gut and salivary glands) and the CNS. Eledoisin-related peptide seems to be roughly equipotent with Substance P at identical ejection currents on the single-cell activity of neurons in this nucleus[2]. Both glutamate and substance P (and its analogue, eledoisin-related peptide) have excitatory effects on the activity of respiratory neurons and reflex interneurons[3].

Twenty-two male albino rats (230-260 g) are anesthetized with chloral hydrate (400 mg/kg, i.p.) and prepared for recording. Briefly, a singlebarrel recording pipette (tip 1 μm) is glued alongside a conventional five-barrel micropipette (tip 15-25/μm) then filled with 2 M NaCI saturated with Fast Green (impedance 4-7 M). The distance between the tip of the recording electrode and that of the five-barrel micropipette is 15-25/zm. Fast Green is ejected at the end of the experiment to identify the recording site. One side barrel of the five-barrel micropipette is loaded with 4 M NaCI for automatic current balancing and the others with three of the following solutions: L-epinephrine bitartrate (0. l M, pH 4.0), L-norepinephrine bitartrate (0.1 M, pH 4.0), Substance P (2.75 mM), physalaemin (2.6 mM), substance P 4-11 octapeptide (3.1 mM), eledoisin-related peptide (20 mM), neurotensin, bradykinin triacetate (15 mM), met-enkephalin (6.5 mM), TRH (48 mM). Spontaneously active cells are recorded in the locus coeruleus or in the nearby mesencephalic nucleus of the fifth nerve whose cells are easily identified by their increased activity upon manipulation of the jaw[2].

在HEK293细胞中表达野生型以及不同突变型SOD1,裂解细胞后添加各抗体(MS785:5 μg/mL, MS27:2 μg/mL, MS785/MS27混合:1 μg/mL)并反应10小时,然后使用Protein G 磁珠进行免疫沉淀。结果显示,MS785和MS27在经过免疫沉淀反应后,未识别出野生型(WT)但与突变体特异性结合。在抗原位点出现突变时,MS785和MS27无法单独识别,但是补充MS785/MS27混合体后,即可检测出来。

◆肌萎缩性侧索硬化(ALS)与SOD1

肌萎缩性侧索硬化(ALS;Amyotrophic Lateral Sclerosis)是一种由于运动神经的细胞死亡导致全身骨骼肌无力、主要症状为肌肉萎缩的神经退行性疾病。90%的ALS为非遗传性发病,10%左右为遗传性发病,并且已经鉴定出了约20种致病基因。其中因氧化还原酶SOD1(Cu/Zn Superoxide Dismutase)的突变导致发病的案例最多,而表达SOD1突变体的转基因小鼠也作为ALS的动物模型使用。借助各种研究手段我们有望阐明由SOD1突变导致的ALS发病机理。

SOD1是一种在活性中心带有铜离子与锌离子的金属酶,主要负责分解细胞内产生的活性氧。虽然SOD1是一种相对较小的蛋白,只有154个氨基酸(人)、分子量仅为24 kDa,但是却有着上百种突变体(图1)。这些突变体基本不会影响分解天然活性氧的酶活性。因此,研究认为,ALS的发病中存在着酶活性以外的其他因素,而伴随突变的毒性获得(toxic gain of function)被视为极有可能的因素。

原本,野生型SOD1形成二聚体,但有关SOD1突变体,东京大学一条秀宪教授等人已澄清了的以下要点:

● 上百种各形各式的SOD1突变体采用了与野生型SOD1不同的共通二级结构(图2)。

● 通过与内质网中的蛋白降解途径(ERAD)的构成蛋白Derlin-1相结合获得细胞毒性。

并且,以SOD1突变体与Derilin-1结合位点为基础,成功开发出能够特异性识别突变体立体结构的大鼠单克隆抗体,克隆MS785和MS27。这些抗体作为阐明SOD1突变引发的ALS发病机制的优秀工具备受期待。

■ 关于克隆MS785与MS27的区别


● MS785是以人SOD1的6~16氨基酸序列,MS27是以人SOD1的30~40氨基酸序列分别作为抗原制备的单抗。

● 均可广泛地检测突变体,但无法检测到某些每个表位均有突变的特定突变体。

● 由于MS785和MS27有互补性,所以混合型抗体可以补全抗原位点突变体的检测。

● Western blot:同时检测突变体与野生型

注意:在变性状态下,不仅会检测到SOD1突变体,也会检测到野生型SOD1。因此,实验过程中必须防止蛋白变性,请勿使用SDS等变性剂。

◆通过免疫染色对SOD1突变体的检测实例


在HEK293细胞表达野生型以及G93A突变体SOD1(FLAG融合蛋白),分别使用MS785、MS27以及MS785/MS27混合型抗体(红)和抗FLAG抗体(绿)进行免疫染色(工作浓度:1 μg/mL)。可以确认,野生型以及G93A突变体均倍倍抗FLAG抗体检测到信号,但是MS785、MS27以及MS785/27混合型抗体均不与野生型反应,仅与突变体反应。

◆缺锌状态下野生型SOD1的检测实例


本抗体可检测出缺锌时野生型SOD1的动态结构变化。不仅ALS研究,也适用于SOD1的基础研究。

● 本产品有在血清缺乏条件以及锌螯合剂TPEN处理下检测出内源性野生型SOD1的检测实例。

● MS785、MS27以及混合物都可用于检测。

锌缺乏和SOD1立体构造改变

OD1是拥有锌离子的金属蛋白。东京大学的一条教授等人明确,在缺锌状态下野生型SOD1会释放锌离子,改变二级结构,并采取SOD1突变体相似的单体结构。这种结构变化,令原本不结合的内质网相关性降解(ERAD)蛋白Derlin-1获得结合能力,诱导了ER 应激(内质网应激)。即使是野生型SOD1,本产品也能识别出缺锌状态下的立体结构变化,检测出缺乏锌的SOD1。因此适用于SOD1基础研究以及ER应激研究。
