WST-3 is a water-soluble tetrazolium dye. In the presence of an electron mediator, WST-3 is reduced to WST-3 formazan by NADH and displays an absorption maxima of 433 nM[1].

体外研究 (In Vitro)
The generally used MTS-based assay is compared with a bioassay employing a water-soluble tetrazolium dye, WST-8, using NFS-60 cells at a concentration of 7×105 cells/mL against 800 IU/mL of PEGylated G-CSF at 24, 48, 72, and 72 h time points to determine the efficacy of PEGylated G-CSF. Further, the optimized WST-8 dye-based assay is used to test the potency of various commercially available PEGylated G-CSF preparations. The results demonstrate enhanced sensitivity of the WST-8-based assay over the conventional MTS-based assay for determining the potency of PEGylated G-CSF using the NFS-60 cell line[1].

WST-1 is a Tetrazolium dye, widely used in the cell viability assays. WST-1 produces a water-soluble formazan product (WST-1 formazan) that occurs at the cell surface in NADPH dependent manner in viable cells[1][2].

WST-1 is used for a wide range of assays that include cell viability, proliferation, plasma membrane electron transport system, superoxide generation, superoxide dismutase, and xanthine oxidase assay[1]. A water-soluble formazan dye is produced from the reduction of WST-1 tetrazolium, of which optical density at 450 nm is measured to evaluate cell viability[1].
