    以 1 mL 工作液为例,取 100 μL 25.0 mg/mL 的澄清 DMSO 储备液加到 400 μL PEG300 中,混合均匀;向上述体系中加入50 μL Tween-80,混合均匀;然后继续加入 450 μL生理盐水定容至 1 mL。

    将 0.9 g 氯化钠,完全溶解于 100 mL ddH₂O 中,得到澄清透明的生理盐水溶液

To demonstrate the feasibility of developing water-soluble NIR dyes, NIR dye-1 (Compound 1h) is further synthesized, which bears several water-soluble groups. As expected, dye NIR dye-1 (Compound 1h) is soluble in pure PBS and newborn calf serum (without addition of any co-organic solvent) with a solubility of at least 2 mM in PBS and 3 mM in newborn calf serum[1].

The porous TiO2 films are immersed in a 0.5 mM D149 (1-material) dye solution in a 1:1 (v/v) mixture of acetonitrile (HPLC) and tert-butanol (LR) overnight once their temperature decreased to approximately 110°C. The samples are then taken out of the dye bath, washed with acetonitrile, and dried. The working electrode and Pt counter electrode [produced using a pre-drilled piece of 2.3 mm FTO glass, coated with one drop of 10 mM platinic acid solution [H2PtCl6] and heated to 400°C for 20 min] are assembled into a sandwich type cell and sealed with a spacer of 25 μm Surlyn. An I-1/I3-1 organic solvent based electrolyte solution [50 mM iodine, 0.6 M 1,2-dimethyl-3-propylimidazelium iodide, 0.1 M lithium iodide in methoxypropionitrile] is introduced via vacuum back-filling. The hole is sealed with a piece of aluminium foil-backed Surlyn[1].

PREX710是一种高体内稳定性的耐光性近红外染料,可用于体内荧光成像、单分子成像、抑制叶绿体自发荧光影响的植物成像以及多重染色。本试剂具有NHS酯结构,可特异性标记氨基。

◆近红外染料在成像中的优点及现有染料的问题


荧光成像是现代生物学中必不可少的技术,目前已研发出各种荧光染料。其大部分为可以被可见光激发的荧光染料,其中蓝色、绿色及红色荧光染料已得到广泛使用。但是,使用可见光作为激发光对生物样品进行荧光成像时,存在以下3个主要的问题:

① 光损害性:由于光能量较高,长时间会对观察对象产生光损害,从而影响实时成像。

② 自发荧光:在可见光激发下,除荧光染料外还可能会观察到生物物质来源的自发荧光。

③ 组织透过性:可见光的组织透过性低,在个体水平的成像中难以观察到组织深处。

 

为解决上述问题,波长比可见光更长的近红外染料(Near Infrared dye; NIR dye)备受期待。近红外染料一般使用700~900 nm范围内的波长作为激发光。长波长光不仅能量低,可抑制光毒性,并且由于生物物质对近红外波长的吸收小,所以自发荧光小,组织渗透性高,而被称为“生物之窗”。传统的近红外染料大部分都以花青素为基本骨架。由于花青素染料系(典型例子:吲哚菁绿)在生物体内的化学稳定性低,且缺乏光稳定性,以及在生物样品中分解快、褪色快,导致在生物样品中的荧光成像不充分。

PREX710作为一种新型的近红外染料,由于使用的是向氧杂蒽骨架引入氧化膦基团的新型骨架,而不是花青素骨架,所以能维持在血液中的化学稳定性,且表现出较高的光稳定性。其有望成为新型的近红外染料,应用于如体内成像等生物样品的长时间成像。

◆特点

◆标记方法概要


本产品为棕色小瓶包装,内含1 mg的PREX710-NHS(绿色),为了稳定NHS酯,以氩气填充。一旦开封,NHS酯可能会逐渐水解,建议在本试剂开封和溶解后立即使用。

1.  小瓶开封后,请立即用DMSO或DMF进行溶解。

1.  注:由于在水中会发生水解反应,不建议溶于水。建议使用脱水级的DMSO或DMF。另外,由于NHS酯在溶液中不稳定,储备溶液请在标记样品前现配现用

1.  因为储备溶液制备后在保存过程中可能会失去标记活性,因此不建议对其进行保存。

2. 将需要标记的含氨基样品溶解于0.1 M的碳酸盐缓冲液 (NaHCO3/Na2CO3,pH 8.3) 。

1.  注:可用磷酸缓冲液PBS代替,但不建议使用含氨基的缓冲剂(如Tris,甘氨酸等),因为NHS可能会与缓冲剂反应而使其失活。

3. 将PREX710-NHS溶液与样品溶液混合,使其至少反应1 h。

4. 通过凝胶过滤或超滤法去除未反应的PREX710染料。

1.  注:染料量及样品使用量可视情况进行优化。

 

标记注意事项

NHS酯与氨基反应时溶液的pH很重要,pH 8.2-8.3最为适合。当pH小于8时,氨基会质子化导致反应效率降低;当pH大于8.5时,NHS酯会发生水解而失去标记活性。

 PREX710的pH稳定性


为了评PREX710的pH稳定性,分别检测了10 h后pH在4.2、7.4、10.3中712 nm处吸光度随时间的变化。在以上的pH条件下,几乎没有观察到吸光度的衰减,由此可推断,本试剂在生物体内的pH范围(pH4~10)中,不受pH影响。

为了评估PREX710的光稳定性,将PREX710溶解于PBS中,使用氙灯(300 W)间歇照射后观察712 nm处吸光度的衰减情况。PREX710即使在氙灯下连续曝光2 h,也完全未观察到吸光度的变化。

为了评估PREX710在血液中的稳定性,将PREX710溶解于FBS(100%)中,观察12 h后在712 nm处的吸光度变化。PREX710在至少12小时内,在血清中的吸光度几乎没有变化,表明在血清中的稳定性高。

 比较蛋白标记过程中的光稳定性


作为标记模型,分别使用PREX710-NHS、花青素系竞争性染料A-NHS以及竞争性染料C-NHS对IgG抗体进行标记,氙灯(300 W)间歇照射后观察光稳定性。在使用了花青素系竞争性染料A及竞争性染料C的标记中可观察到吸光度迅速衰减,但PREX710即使在IgG抗体标记状态下,吸光度在长时间的光照射下也几乎不发生变化。

◆应用数据


■ 小鼠的体内血管成像


制备使用PREX710-NHS标记的氨基葡聚糖(平均分子量为70,000),尾静脉给药于4周龄的小鼠,使用共聚焦激光显微镜(激发/荧光波长=638 nm/667~733 nm)对使用开颅法处理的脑血管进行深部成像。PREX710在血液中可长时间稳定,由于它可在不易受血红蛋白自发荧光影响的波长下激发,成功地进行了清晰的脑血管深部成像,解决了血液成像的一大难题。

■ 植物成像


使用经PREX710-NHS标记的八聚精氨酸R8(PREX710-R8),对小立碗藓(Physcomitrella patens)的原生质体细胞壁进行染色,并用荧光显微镜观察。在703~717 nm/754~816 nm的激发/荧光波长下观察PREX710-R8,检测到在300~400 nm/>420 nm的激发/荧光波长下叶绿素为自发荧光。通过使用PREX710可以区分叶绿体的自发荧光,从而检测到清晰的信号。由于PREX710不易受叶绿体的自发荧光影响,适用于植物成像。

■ 使用PREX710的单分子成像


构建单分子成像的模型,分别制备经PREX710-NHS和花青素系竞争性染料A-NHS标记的抗生物素蛋白,随后通过生物素偶联固定在载玻片上。点上的信号是由单个抗生物素分子引起的荧光信号。以10帧/秒的采集速度,120 s内连续追踪单分子的信号。观察到花青素系竞争性染料A标记的各个信号会逐渐衰减,到40 s后荧光信号已几乎完全消失;而经PREX710标记的各个点信号即使在单分子水平下也能连续观察120 s,并只观察到了轻微的衰减。
