(In Vitro)

A first characterization and comparison is done by developing an easy and direct measurement method based on a pH indicator system using Bromothymol Blue (BTB) as the indicator and potassium phosphate as the buffer. A pH-shift assay is developed on the basis of BTB as the pH indicator and potassium phosphate as the buffer component[1]. Three pH indicators are tested for the direct determination of 2- 2-keto-L-gulonic acid (2-KLG) production on a plate. The results show that Bromothymol Blue is superior to the other two indicators in terms of the obvious color change and a suitable pH range (blue to yellow at pH 6.5-7.5). Upon the addition of a Bromothymol Blue solution (0.1%, w/v) to an agar plate, zones surrounding colonies of K. vulgare 07 mutants change their color from blue to yellow because K. vulgare 07 mutants release 2-KLG on agar plates, thereby acidifying surrounding areas around colonies[2].

For all three enzymes, an expression in the 96-deep well scale is performed. Therefore, electrocompetent E. coli ArcticExpress(DE3) cells are transformed with the corresponding plasmid. Single clones are picked using the CP7200 Colony Picker and transferred to 96-deep well plates filled with 1.2 mL autoinduction media by a MicroFlo Select dispenser. After incubation (36 h, 37°C at 1,000 rpm), further processing is done manually. First, 100 μL of cell culture is transferred into a 96-well plate (U-shaped bottom) and harvested by centrifugation (4,570 rpm, 10 min at RT) while the supernatant is discarded. The frozen pellets (1 h at -20°C) are thawed at room temperature for one hour to improve cell lysis. Lysis is continued by the addition of 30 μL lysis buffer (3 h, 1,000 rpm, 37°C) containing 2 mM KPi, pH 7.0, 2 mM MgCl2, 10 μg/mL DNaseI, 100 μg/mL lysozyme. Next, 120 μL buffer (2 mM KPi, pH 7.0) is added followed by centrifugation (3,000 rpm, 15 min at RT). For the photometric measurement, 20 μL of the crude extract is transferred to a 96-well plate (F-shaped bottom) and the reaction is started by adding 180 μL master mix to give a final volume of 200 μL (2.5 mM KPi, pH 7.0, 2 mM MgCl2, 25 μg/mL BTB and 5 mM keto-deoxy-D-glucarate). The measurements are carried out for 60 min at 2-min intervals. Depending on the enzyme, different time windows are used for the activity calculation[1].

CytoSeeing是一种新型细胞染色试剂,只需添加到培养基即可迅速对细胞质进行染色。荧光观察细胞质后,只需更换不含CytoSeeing的培养基,便可轻松去除细胞内的荧光染色。

◆关于CytoSeeing

已知细胞核与细胞质的形态变化与细胞分化、功能、信号反应相关。传统的细胞质染色探针,主要作为细胞示踪剂使用,一旦摄入细胞,即使去除培养基中的试剂也会残留在细胞内。随后,细胞内的染色试剂随着细胞分裂的过程逐渐褪色,通常在3~6代左右便会消失。但是如果染色试剂残留在细胞内的话,实时成像观察细胞质与细胞核的形态后,难以立即使用其他探针进行成像。

不同于传统试剂,CytoSeeing是一种仅对细胞质进行染色且可清洗的细胞染色试剂。

◆特点


● 只需添加到培养基即可对细胞质进行染色。

操作方法概要

1. 取适量细胞进行培养;

3. 在适合细胞的温度下孵育几分钟;

Nile blue A is a basic oxazine dye which is soluble in water and ethyl alcohol. Nile blue A is a satisfactory stain for PHB granules in bacteria and is in fact superior to Sudan black B for this purpose. Poly-p3-hydroxybutyrate granules exhibits a strong orange fluorescence when stained with Nile blue A. Nile blue A appears to stain many more PHB granules than Sudan black B does and is not as easily ished from the cell by decolorization procedures[1]. Nile blue A is used as a stain for polyhydroxyalkanoic acid-accumulating microorganisms or to detect polyhydroxyalkanoic acids in microorganisms. Escherichia coli cells that do not accumulate detectable polyhydroxyalkanoic acids can be stained with Nile blue A and that this staining is sufficient for identifying these cells in fluorescence-activated cell sorting (FACS) experiments. Nile blue A staining does not affect either surface display of peptides or specific labeling of these peptides by a second fluorescence. Staining E. coli for flow cytometry using Nile blue A is an easy-to-handle and low-cost alternative to other fluorescent dyes or the intracellular expression of, for example, green fluorescent protein[2]. Nile blue A is one of the most studied benzophenoxazine dyes, as a potent photosensitizer for photodynamic therapy. The dye when administered intravenously disperses throughout the body by circulating through blood and is taken up by most cells that emphasize its interaction with various biomolecule[3].

Cell Assay

1% aqueous solution of Nile blue A is prepared and filtered before use. Mild heating may be necessary to fully dissolve the stain. Heat-fixed smears of bacterial cells are stained with the Nile blue A solution at 55°C for 10 min in a coplin staining jar. After being stained, the slides are washed with tap water to remove excess stain and with 8% aqueous acetic acid for 1 min. The stained smear is washed and blotted dry with bibulous paper, remoistened with tap water, and covered with a no. 1 glass cover slip. The preparation is examined with a Nikon Labphot microscope with an episcopic fluorescence attachment[1]. The PHA− strain Escherichia coli UT5600(DE3) is stained with Nile blue A. In an Erlenmeyer flask, 20 mL of Luria–Bertani (LB) broth is inoculated with one colony of UT5600(DE3) and incubated for 14 h at 37 °C and 200 rpm. Subsequently, 20 mL of LB broth containing Nile blue A in a final concentration of 0.5 μg/mL is inoculated with 200 μL of the 14-h culture and cultured to an optical density at 578 nm (OD578) of 0.6. As a control, 20 mL of LB broth (without Nile blue A) is inoculated with 200 μL of the 14-h culture and is also cultured to an optical density at OD578 of 0.6. Every 20 min, the OD578 is determined for both cultures to verify whether there is any influence of the dye on the growth of the bacteria[2]. Nile blue A stock solution is prepared using ethanol as the solvent. The concentration of NB is maintained at 5 μM for all the studies. The solutions are left for 1 h to achieve equilibrium before spectral measurements. The absorption spectra are recorded using Shimadzu Spectrophotometer (UV-1800) and the emission spectra are recorded using Jobin–Yvon Spectrofluorimeter. A 450 nm nano-LED is used as the light source and the fluorescence lifetime is collected at λem=672 nm[3].

Calcein Blue, a membrane-impermeant fluorescent dye, is a coumarin derivative that contains an iminodiacetic acid structure. Calcein Blue is also a metallofluorochromic indicator[1][2][3].

(In Vitro)

The fluorescence of Calcein Blue (CB) is quenched by the Fe2+ ion in the Calcein Blue-Fe2+ complex. When dopamine is added to a solution of the Calcein Blue-Fe2+ complex, a dopamine-Fe2+ complex is formed as the result of a ligand exchange reaction between Calcein Blue and dopamine which permits the fluorescence from Calcein Blue to be recovered. The fluorescence intensity at the wavelength of 440 nm (at the excitation wavelength of 340 nm) is found to be proportional to the concentration of the dopamine added to the Calcein Blue-Fe2+ complex solution, which permits dopamine to be quantitatively determined. The Calcein Blue-Fe2+ complex shows a high selectivity for dopamine among the catecholamines and related compounds examined[1].

Cell Assay

The recommended final working concentrations is usually between 1 and 10 μM to minimize potential artifacts. Generally, 15 minutes to 1 hour is sufficient for cellular uptake and processing of the dyes. A stock concentration of 5 mM 4-methylumbelliferone-8- methyliminodiacetic acid, commonly known as calcein blue is prepared in 0.1 M potassium hydroxide, and the pH is neutralized using 0.1 N hydrochloric acid. The calcein blue stock prepared is diluted to 200 μM (working stock) in DPBS and stored for further use. A concentration of 10 μM calcein blue is prepared from the working stock by diluting in DPBS. From this, 200 μL is added to a 96-well plate and the excitation/emission (Ex./Em.) maximum is determined using the micro-plate reader[1].
