BODIPY-cholesterol assay
To prepare a stock solution of BODIPY-cholesterol for this assay, the analogue is reconstituted in DMSO to a stock concentration of 1-10 μM[1].
Cells diluted with PBS (or other buffers) are washed (300×g, 10 min) to remove excess diluent and the loose pellet resuspended with vehicle media to a proper concentration[1].
Cell suspensions are labeled with BODIPY-cholesterol (final concentration of 0.5-5 µM), mixed thoroughly and incubated for 10 -20 mins at 37 °C[1].
The cells are diluted with each of the capacitation media conditions as outlined in Incubation media to a final concentration and incubated for 2 h prior to flow cytometric assessment[1].

Ingenol is a PKC activator, with a Ki of 30 μM. Ingenol induces ornithine decarboxylase activity (1, 3 mM), and causes morphological changes (1 mM) in primary mouse epidermal keratinocytes. Ingenol (125 μM, 250 μM, 500 μM, 1 mM) also inhibits cell-cell communication[1].

TAPl-/- or C57BL/6 macrophages are co-incubated with either bacteria or polystyrene beads containing the 257-264 epitope from ovalbumin [OVA Peptide(257-264)], which binds the mouse class I molecule Kb. The source of the OVA(257-264) epitope is either the Crl-OVA(257-264) (Crl-OVA) fusion protein, the maltose binding protein (MBP)-Crl-OVA fusion protein, native OVA or bacterial recombinant OVA (rOVA); Crl-OVA, MBP-Crl-OVA and rOVA are each expressed in bacteria, and Crl-OVA and MBP-Crl-OVA purified from bacterial lysates and native egg OVA are coated onto polystyrene beads[1].

The standard reaction mixture contains 1×l0-7 M MCLA hydrochloride (MCLA), 5×10-5 M hypoxanthine, XOD (6.5 U), SOD (0.2 to 20 ng/mL) or none, and 50 mM Tris-HCl buffer containing 0.1 mM EDTA at pH 7.8, in a total volume of 3.0 mL. Chemiluminescence measurement is initiated by the addition of MCLA hydrochloride to the standard incubation mixture excluding XOD, continued for 4 min without additive and for an additional 4 min after the addition of XOD. Chemiluminescence is measured using a luminescence reader at 25°C[1].

Isoquercitrin (Isoquercitroside) is an effective antioxidant and an eosinophilic inflammation suppressor.

(In Vitro)

Isoquercitrin occurs widely in plants.10 μM of Isoquercitrin of the seven compounds partially but significantly blunt the negative effect of H2O2 on RGC-5 cells, as the viability of cells in the presence of H2O2 is increased from approximately 63% to 83% and 90% at 10 and 50 μM, respectively. Indeed, 50 μM Isoquercitrin is more effective as a neuroprotectant than the same concentration of EGCG[1].

(In Vivo)

In animals receiving Isoquercitrin, eosinophil counts are lower in the BALF, blood and lung parenchyma. Neutrophil counts in blood and IL-5 levels in lung homogenate are lower only in Isoquercitrin-treated mice. No alterations in mononuclear cell numbers were observed[2].

Animal Administration

Mice[2]
BALB/c mice are immunized (ovalbumin/aluminum hydroxide, s.c.), followed by two intranasal ovalbumin challenges. From day 18 to day 22 after the first immunization, the mice receive daily gavages of Isoquercitrin (15 mg/kg) or quercetin (10 mg/kg). Dexamethasone (1 mg/kg, s. c.) is administered as a positive control. Leucocytes are analyzed in bronchoalveolar lavage fluid (BALF), blood and pulmonary parenchyma at 24 h after the last ovalbumin challenge. Interleukin-5 (IL-5) is analyzed in BALF and lung homogenates[2].
